State Key Laboratory of Medical Molecular Biology [1] Relying on the Institute of Basic Medicine of the Chinese Academy of Medical Sciences, the laboratory was established under the guidance of the national long-term scientific and technological development plan and in combination with the characteristics of medical research. As it is located in the central area of Beijing, it is also associated with the library of the Chinese Academy of Medical Sciences, which has a rich collection of books and a long history, and the internationally famous large general hospital—— Peking Union Medical College Hospital Adjacent to each other, it has a special and excellent environment for the development of basic medical research.
The State Key Laboratory of Medical Molecular Biology was invested about 2 million yuan by the Chinese Academy of Medical Sciences and rebuilt on the original site of the Institute of Zoology, covering an area of about 1500 square meters. It passed the expert argumentation on February 25, 1991, and in April 1992, the Ministry of Health issued a document to officially approve the implementation of the establishment and opening up of the system; In November 1993, it passed the acceptance organized by the State Planning Commission; In 1996 and 2001, it passed the expert evaluation organized by the National Science and Technology Commission twice, and both of its achievements were among the best in Level B.
After the completion of the new scientific research and teaching building of Peking Union Medical University in 2003, the key room was moved in, covering an area of 3800 square meters, making it more of a modern laboratory, providing a good scientific research environment for carrying out basic research in medical molecular biology and receiving advanced studies and open topics from international researchers, It provides a guarantee for the laboratory to better complete the tasks of undertaking major international scientific research projects and cultivating talents.
The foundation of this laboratory is the original Peking Union Medical College (Peking Union Medical College) Department of Biochemistry has been engaged in molecular biology research in protein, nucleic acid and other related important fields for more than 40 years since the early 1950s. It has accumulated rich experience and trained a large number of research talents. In recent years, with the support of relevant departments, we have actively carried out international and domestic academic exchanges and cooperation, and gradually formed a research team with advanced equipment, strong technical force, advanced biological technology and methods, and young and middle-aged scientists as the backbone under the support of domestic and foreign multi-channel funds. There are 25 researchers and associate researchers (fixed staff) in this room, including 3 academicians of the Chinese Academy of Sciences, 2 academicians of the Chinese Academy of Engineering, and 18 doctoral supervisors. It has formed its own research characteristics, has high-level academic leaders in various research directions, and has become a leading laboratory in China and has a certain influence in the world.
The laboratory implements the director responsibility system, and the current director is Academician Liu Depei. The laboratory implements the evaluation system of academic committee, which is responsible for the examination and approval of laboratory research direction, construction planning, research topics, achievement evaluation and fund use principles. Two thirds of the members of the Academic Committee are outdoor experts with a term of office of three years. The current director of the Academic Committee is academician John Boqin.

The State Key Laboratory of Medical Molecular Biology of the Institute of Basic Medicine of the Chinese Academy of Medical Sciences is a comprehensive open laboratory in the field of medical research in China. This research group is an important part of the State Key Laboratory. Academician Liu Depei, the academic leader, and Academician Shen Yan, the research backbone, have been engaged in medical research together in this laboratory for nearly 20 years, After studying abroad and making great achievements in scientific research, Professor Zhu Dahai and Professor Zhang Xue joined the research group as distinguished professors to jointly carry out research work in important research directions of the laboratory. The researchers of this group have made important research results in the screening of disease genes and the regulation of eukaryotic gene expression. They have been rated as "outstanding contribution talents" at the national/provincial/ministerial level for many times, and have won many national/provincial/ministerial science and technology awards. He has made outstanding achievements in the function and pathogenesis of disease related genes, and has won the support of the National Science Fund for Distinguished Young Scholars.
With the development of molecular biology, especially genomics and Proteomics With the further development of, various physical/chemical and biological new methods and technologies are constantly emerging and applied, generating and accumulating a large amount of genetic information. On the basis of existing work, this research group will further focus on outstanding talents and apply the combination of micro and macro systems biology Integrating innovative thinking and high-tech platform, aiming at the key scientific problem of the function and expression regulation of important genes related to the occurrence/development of major diseases, and deeply exploring its internal mechanism. Through our efforts, we hope to make new breakthroughs in disease genomics research and apply the results of basic research to medicine, laying a theoretical foundation for gene diagnosis and treatment of major diseases.

Proteomics aims to study the characteristics and functions of proteins, so as to comprehensively understand the growth process of normal and abnormal cells, the interaction between proteins and the regulation mechanism of protein level. High speed or Ultrafast freezing centrifuge It can effectively separate and purify proteins; Liquid chromatography can analyze and prepare samples; Rapid electrophoresis and two-dimensional electrophoresis are standard techniques for proteome separation; senior Gel imager After electrophoresis, the separated proteins will be stained and imaged on the gel and further analyzed.
Core technology: HPLC is a classic instrument analysis method, but it has not been fully used in the field of nucleic acid analysis. The reason is that in many cases, nucleic acid analysis needs to be carried out under high temperature denaturation conditions (52-80 ℃), while the traditional HPLC separation system cannot solve the problem of high temperature resistance. The best Si matrix column can only withstand 60 ℃ at most, and its life is very short at this temperature, The unique DNASep column developed by American Transgenic Company is the core part of WAVE nucleotide fragment analysis system, DNASep column uses PS/DVB polymer microsphere as the stationary phase matrix and chemically bonded stationary phase C18. Thus, the column filler formed by PS/DVB and C18 has good thermal stability. It is the characteristics of high temperature resistance that ensure the accuracy and repeatability of its analysis results. At the same time, this separation column system is acid and alkali resistant, with long service life and low analysis cost. Because such polymer microspheres are small in size and have a large specific surface area, which ensures more interaction opportunities between nucleic acid fragments and stationary phase, the separation efficiency is high and the analysis time is short.
Instrument and equipment: AKTA explorer chromatography systems, WAVE denaturing high-performance liquid chromatography (DHPLC), micro purification liquid chromatography system, fast protein electrophoresis system Full automatic gel imaging analysis system
