Operation method of anaerobic incubator:
1. Check and close the door in the sampling room;
2. Open the door outside the sampling room, put the bacteria into the sampling room, and then close the door;
3. Three nitrogen replacement processes in the sampling room: open the vacuum pump, pump the vacuum above 500ml Hg (66kPa) and stop, then manually open the nitrogen valve 2 for air intake, make the pointer return to zero, and close the valve 2. Repeat the operation for the second time. In the third operation, stop the vacuum above 500ml Hg (66kPa), then open valve 4 to feed the mixture, return the pointer to zero, and close valve 4. The three nitrogen replacement processes in the sampling room are completed;
4. If the selected vacuum degree is low, the replacement times need to be increased;
5. Open and close the door outside the sampling room, and then pump 100ml Hg (13kPa) at low vacuum for inspection;
6. Adjustment of output pressure of mixing gas cylinder and nitrogen cylinder: adjust the pressure reducing valve to make the output pressure about 0.1Mpa.
Conditions for long-term use of anaerobic incubator:
1. Open the Meilan indicator paper in the operation room every day for observation. If it is abnormal, it must be re ventilated;
2. It is necessary to continuously input a small amount of mixed gas for a long time, so that the added nitrogen can be combined with a small amount of oxygen through catalytic absorption to ensure the indoor anaerobic state, and the flow rate of the added mixed gas is about 10ml per minute;
3. Run the culture continuously for one day, and replace the deoxidizer and desiccant once.
The use process of anaerobic incubator is actually two steps: the formation of anaerobic environment in the operating room and the placement and cultivation of bacteria, but each step requires many small steps to complete. Let's take a look at it together.
1、 Formation of anaerobic environment in operation room:
Place the necessary accessories and appliances according to the use requirements, and put two five toxic plastic bags into the operation room. After the equipment is turned on, press the main power button on the control board to power on the equipment. First, insert a rubber tube into the air inlet of the operating room, and the other into a plastic bag, connect the nitrogen air inlet circuit, open the nitrogen control valve of the operating room of the anaerobic incubator, make two plastic bags full of nitrogen, close the solenoid valve, then tighten the bag mouth, put latex gloves on the flange ring of the observation plate and tighten them, and gradually put the nitrogen in the plastic bag into the operating room until it is completely prevented. In the operating room of the anaerobic incubator, turn on the palladium particle deoxidizer, connect the power supply of the deoxidizer catalyst for catalytic deoxidization, and drop a little clean water on the reaction area of the anaerobic indicator strip. After 1-2 hours, observe its change. When the reaction part turns dark blue to light blue, the operating room reaches the anaerobic environment.
2、 Insertion and culture of strain:
Check whether the door in the sampling room of the anaerobic incubator is closed tightly, open the door outside the sampling room, close the outside door after putting the bacteria into the sampling room, fill the sampling room with nitrogen for three times, then open the oxygen valve in the sampling room to let the pointer return to zero, and close the valve. Anaerobic incubator needs the condition of long-term continuous use. Put an indicator strip in the operation room every day for observation. If it is not normal, it must be re ventilated. It is necessary to continuously input a small amount of mixed gas for a long time, so that the supplemented nitrogen can combine with a small amount of nitrogen through catalytic absorption, ensuring the anaerobic state in the room. The flow rate of the supplemented mixed gas is selected to be about 10 ml per minute, After continuous cultivation for one day, replace the deoxidizer and desiccant once.
