过渡金属催化的不对称氢化反应是制备多种对映体纯化合物的最有效方法之一。通过设计新的配体来发现具有改进效用、活性和选择性的催化剂的可能性在这一研究领域中起着至关重要的作用。因此,用于催化不对称氢化反应的新型手性配体数量迅速增长。在这种情况下,手性二膦(例如双极管、BINAP、DIOP、DuPhos)是最有用和最通用的配体(Lin等。, 2001). 与手性芳基膦配体在Rh-和Ru-催化的不对称反应中的巨大成功相比,含有杂环部分(例如吡啶基)的手性膦配基还相对未被探索。

最近,我们开发了一种手性联吡啶膦配体,即2,2'6,6'-四甲氧基-4,4'-双(二苯基膦)-3,3'-联吡啶,(II)(P-Phos;Pai等。,2000),发现其在2-(6'-甲氧基-2'-萘基)丙烯酸和β-酮酯(Pai等。, 2000).

P-Phos合成的关键步骤是外消旋体(II)与光学活性(+)-二苯甲酰基的光学拆分-L(左)-酒石酸[(+)-DBT],(III),作为分解试剂(Takaya等。1988年),这是非常有效和高度选择性的。经过几次试验和我们的实验修改,光学纯的二吡啶基膦氧化物()-(二) 和(S)-(II)以高对映体纯度获得。因此,当(2)的解S公司将乙酸乙酯中的,3S)-(+)-DBT添加到二氯甲烷外消旋体(II)的沸腾溶液中,(-)-(II)和(+)-DBT[(-)–(II)-(+)-DBT,(I)]的1:1络合物沉淀为结晶固体。根据与(2)的内部比较S公司,3S)-(+)-DBT,确定(-)-II的绝对构型为S。

(I)(图1)的晶体结构在晶格中还包含无序氯仿分子。两个吡啶环的最小二乘平面之间的二面角为75.9(0.1)°。晶体由无限链组成,其中等摩尔量的(-)-(II)和(+)-DBT通过P的O9和O14之间形成的两个分子间氢键交替连接(-)-(II)的O基团和(+)-DBT的COOH基团的H原子。O6··O9和O4··O14ii(ii)距离分别为2.551(2)和2.554(2)Ω[对称码:(i)1/2+x个, 3/2 −, 1 −z(z); (ii)1−x个, −1/2 +, 3/2 −z(z); 图2和表1]。(I)的低溶解度归因于观察到的聚合物结构。

至外消旋体(II)(Pai)的沸腾溶液等。(1.87 g,2.76 mmol)存于氯仿(28 ml)中,添加(-)-DBT,(II)存于乙酸乙酯(28 ml)中的溶液。将混合物回流,直到形成白色固体,然后在室温下放置一夜。在室温下真空干燥固体产品,得到1:1的配合物()-(二) 和(-)-DBT(93%产率)。将含有(S)-(II)-(+)-DBT复合物的母液浓缩至干燥,得到固体物质。将其溶解在氯仿(10 ml)中,然后用10%的氢氧化钠水溶液(10 ml)处理。用两份氯仿(25 ml)提取混合物。蒸发溶剂得到粗(S)-(II),将其溶解在回流氯仿(15 ml)中,并向该溶液中添加乙酸乙酯(20 ml)中的(+)-DBT溶液。在回流温度下搅拌混合物,直到形成白色沉淀,然后在室温下放置一夜。干燥白色沉淀,得到纯(S)-(II)-(+)-DBT络合物(92%产率)。用于数据收集的晶体是在室温下从氯仿-乙酸乙酯(1:1)饱和溶液中缓慢蒸发获得的。
