皮质类固醇是一类能够发挥多种生理和代谢活性的化合物;特别是,糖皮质激素可以影响脂质、碳水化合物和蛋白质的代谢,影响电解质平衡(Goodman&Gilman,1993)。标题化合物甲泼尼松(I)是一种类固醇抗炎药,适用于风湿、胶原蛋白和皮肤病,其抗炎效力相当于可的松的六倍。美泼尼松,16岁β-泼尼松的甲基类似物(Δ1可的松或17α,21-二羟基-孕二醇-1,4-二烯-3,11,20-三酮),由于观察到甲基取代甾体D环会改变(减少)钠+化合物的保留特性(劳瑟等。, 1966). 泼尼松(II)的结构已通过单晶X射线晶体学测定,数据已保存在剑桥结构数据库(CSD,Allen&Kennard,1993)中,参考代码为PRGDOL(Tseikinsky等。,1979年)。目前的工作是一项正在进行的研究的一部分,旨在确定生物活性化合物(Vega)的三维结构等。,2001年),关于(I)的报告。

化合物(I)在空间群中结晶P(P)21,在不对称单元中有两个独立的分子(见图1)。在类固醇核中,A环是平面的,而其他六元环明显偏离平面性,并采用接近椅子的构象。Cremer&Pople(1975)关于分子(I)A和(I)B的环折叠参数如下:对于环B,QT型为0.547(4)和0.562(3)Ω,θ2为174.9(4)和176.8(3),以及ϕ2为15(4)和126(7)°,对于环C,QT型为0.589(4)和0.583(4)Ω,θ2为170.4(3)和177.8(3),以及ϕ2为58(2)和68(9)°。在这四个环中,不对称参数表明存在三个镜面和一个典型的理想椅子构造(Duax等。, 1976). 五元D环位于13β, 14α具有q的折叠参数的半椅构象2=0.454(4)和0.484(4)Ωϕ2=11.9(4)和12.8(4)°[分别为(I)A和(I)B],而不对称参数为DS公司(C13)=0.075(2)和0.085(2)以及D2(C13–C14)=(I)A和(I)B分别为0.026(2)和0.024(2)。

(I)A在(I)B上的分子叠加,使用XP(极限编程)(Sheldrick,1991)给出了0.30Ω的均方根偏差。如图2所示,分子在原子O3(O3-O3B偏差为0.791º)、O11(0.479º)和O21(0.459º)的位置上差异很大。在(I)B中,A环的平均平面与最小平方C5··C17基准面(Duax等。, 1976); 然而,在(I)A中,二面角为30.95(8)°,因此O3B位于加利福尼亚州与O3A相比,距离C5··C17基准面1º[2.708(3)1.743 (2) Å]. 这种差异可能是O3原子参与的分子间相互作用的结果。施主···受体距离和氢键角度相差0.25°和25°(表2),从而使O3B相互作用比O3A更强。在(II)中,O3原子与C原子弱接触(D··a=3.379º,H··a=2.615°和D-H··A126°)。因此,分子(I)A显示的几何结构可能是由于O 3A氢键相互作用的最大化,即施主··受体距离的减小,以及A环的伴随位移(另见图3)。C11平面(C9-C11-C12-O11)相对于(I)B中椅子(C8-C9-C12-C13)主平面的偏差比(I)A中的大2°,二面角分别为52.66(13)°和50.61(16)°。弱的C1··O11和C19··O1氢键可能会产生影响,但这种情况似乎不太可能发生,因为这两个独立分子中的氢键具有可比性(表2)。因此,由相应的a环引起的远距离效应可能是最合理的解释。C20平面(C17-C20-O20-C21)相对于C5-C17参考平面的方向在两个独立分子中非常一致,二面角的值分别为70.9(1)°和70.5(1)℃[(I)A和(I)B,保留];因此,O17A和O17B与C20平面的距离分别为1.593(2)Å和1.752(3)Å。关于C20平面,如扭转角O20-C20-C21-C21-O21所示,O21的方向为正同周面[6.2(5)°和3.8(5))°(对于(I)A和(I)B)]。从表2可以明显看出,与O21A不同,O21B参与与O20B的分子内相互作用,从而形成五元环。

中(I)和(II)的比较XP(极限编程)表明后者更接近于(I)B而不是(I)A,均方根误差分别为0.14º和0.28º。在(I)B和(II)之间的叠加中,在O20-C20-C21-O21链内遇到了主要差异,即原子O20(0.319Å)、C21(0.255Å)和O21(0.248Å)。然而,在与(I)A的叠加中,原子O3(0.824º)、O20(0.452º)和O11(0.388º)出现了主要偏差。与(II)一样,O20相对于C16-C17键为正向斜,C16-C17-C20-O20扭转角分别为44.9(4)、47.9(2)和33.2°[(I)A、(I)B和II]。注意,O20在(I)中比在(II)中离C16远,这可能是由于甲基成员在O原子上引入空间位阻的结果。非常弱的C-H··O氢键会产生影响。C18··O20相互作用可视为吸引接触(表2),而由于弯曲几何形状,C22··O2相互作用(平均D··A=2.872º,H··A=2.609º和D-H··A=96°)可视为排斥接触,因此净效应是(I)中O20··C16距离的扩大。在(II)中,有一个非常弱的C16··O20氢键(D··a=2.855º,H··a=2.404º,D-H··a=107°),这在(I)中没有观察到。

类固醇核的纵向扭曲,由假扭曲角C19-C10··C13-C18(Duax等。,1976),在两个独立分子中相似[(I)A和(I)B分别为2.6(3)°和2.7(3)],并且该角度略小于(II)[4.6°]中的角度。此外,O17和C18之间的相互取向在三种结构中非常一致,如(I)A、(I)B和(II)的扭转角C18-C13-C17-O17[164.6(2)、166.7(4)和165.4°]的值所示。(I)晶体中的结构内聚是通过O-H··O型氢键实现的。如图3所示,链由两个相互连接的独立分子形成通过O21B···O3A氢键,并由两个分子和翻译邻居通过O17A··O3B(x个− 1,+ 1,z(z)−1)氢键。链由O21A··O17A传播(-x个,+ 1/2, −z(z))和O17B··O21B(-x个+ 1,− 1/2, −z(z)+1)通过晶体双轴的氢键。传播方向几乎垂直于类固醇的最大尺寸,因此链排列成平行于b条轴。
