合成和结晶
在钠和二苯甲酮上干燥甲苯。所有其他溶剂和试剂都是从Alfa Aesar或Fisher Scientific收到的,未经进一步纯化。已报道4-氰基-3,5-双苯基吡唑的合成(Som,2013)). 本工作对程序进行了修改,以使最终产品再结晶。将0.69克(17.24毫摩尔,60%分散在矿物油中)氢化钠加入100毫升干燥甲苯中。向该溶液中添加2.50 g苯甲酰乙腈(17.24 mmol),立即出现气泡。将混合物搅拌18 h,然后添加苯甲酰氯(2.43 g,17.24 mmol)。将反应混合物搅拌过夜,然后使用100 ml 0.2进行三次提取M(M)每次NaOH溶液。将水层与HCl/H结合并酸化2O(50/50)溶液至pH~1。立即出现白色沉淀,并用三份100 ml乙酸乙酯萃取。减压下去除溶剂得到2.55 g(10.24 mmol,产率59.40%)粗产物,粗产物从乙醇中重结晶得到2-氰基-1,3-二苯基-1,3-丙二酮(1.38 g,5.54 mmol,32.15%)。然后将该二酮化合物(1.38克,5.54毫摩尔)与肼一水合物(0.28克,5.54毫摩尔)在100毫升甲醇中反应并搅拌过夜。在减压下除去溶剂,得到淡黄色固体的4-氰基-3,5-双苯基吡唑粗产物,其从乙醇(1.01克,4.65毫摩尔,27.00%)中重结晶。红外光谱显示特征峰位于3177厘米−1和2227厘米−1N-H和C≡N分别延伸。这个1H NMR(氯仿-d日)在8.06显示化学位移(d日,4H),7.64(吨和下午7.54点(吨,2H)。
